Did Freeza power up twice in his second form?

The second time (before Piccolo removed his weights), was it really a power up or did he just get more serious
in effort?

I think he probably did a power up, since he wasn't able to damage Piccolo with a full power blast earlier. A lack of a visible power up scene doesn't necessarily mean he didn't power up, seeing as Cell does the same thing when he goes from being weaker than Super Vegeta to superior to him on a whim.
