How would this fight go down if both were serious? Does Luffy far surpass this level after his training in prison?

He most likely will be above it via general scaling and story structure, though by how much is debatable. Until we get further feats to reinforce it, I'd say Luffy's current gains are just modest enough to win with high-diff as long as Boundman or Snakeman don't run out in time, which should be possible given how he managed against someone with far better feats like Katakuri despite also taking an off guard spear to the side.

Well didn't he handle hits from Big Mom much better than Queen did?

He did, though that can be chalked up to his plot armour naturally top tier durability. In a battle of attrition he'd certainly win, but that's assuming Queen allows him to keep up something so advantageous.
